Minister of SMEs and Startups Park Young-sun has tendered her resignation.
The move was announced in a text message to reporters on Wednesday.
Earlier, the minister thanked them for their support during her past year and nine months in office.
Her last official duty will be chairing a staff meeting at 2 p.m.
Park is expected to formally declare her bid to run for Seoul mayor in the April by-elections.
The presidential office also announced a partial Cabinet reshuffle, including the nomination of Kwon Chil-seung, a ruling Democratic Party lawmaker, to succeed Park.
