People traveling to Australia from most countries will need to test negative for COVID-19 within three days of their departure.
Health Minister Greg Hunt said the requirement will come into effect at midnight Friday, local time.
All international passengers will also have to wear masks on their flights as part of tightened measures to prevent the potential spread of virus.
Only New Zealand and a handful of Pacific Island nations will be exempt from the new rules.
