Samsung Electronics reported on Monday two coronavirus infections at its Gwangju Campus, some 300 kilometers south of Seoul.
The company said it was informed by the local health office that two workers from its refrigerator production line tested positive for the disease.
The assembly plant has been shut down until Wednesday.
Authorities are looking into the route of infection as well as trying to determine who the workers came into contact with.
The tech giant has reported dozens of cases at other sites and offices, including at semiconductor plants, since the start of the pandemic.
Kia Motors also suspended two of its three plants in Gwangju earlier in the day after four line workers tested positive for COVID-19.
These latest outbreaks to hit major industrial plants come amid mounting concerns about production losses this year.
